兔骨髓间充质干细胞穴位移植与静脉移植对梗死区类心肌细胞新生影响的比较

摘    要:目的:探讨急性心肌梗死后0天与7天穴位与静脉分别注射骨髓间充质干细胞(MSCs)在梗死区分化为类心肌细胞的情况。方法:用开胸结扎冠状动脉前降支的方法建立急性心肌梗死模型,将已建立急性心肌梗死模型的日本大耳白兔随机分为5组,抽取健康日本大耳白兔骨髓,分离培养MSCs并传代,于急性心肌梗死形成后0天与7天穴位及静脉注射干细胞。免疫组化brdu染色,取有brdu阳性细胞的切片进行免疫组化染色,显微镜下观察心肌特异性肌钙蛋白I染色。结果:模型对照组心肌梗死区未见brdu标记的MSCs;穴位注射干细胞0天组brdu阳性细胞的平均光密度值较静脉注射干细胞0天组增高趋势明显P<0.05;穴位注射干细胞7天组brdu阳性细胞的平均光密度值较静脉注射干细胞7天组增高无明显差异P>0.05。结论:在0天穴位与静脉注射MSCs迁移至梗死区,在梗死区域内存活并分化为类心肌样细胞;在7天穴位与静脉注射MSCs迁移至梗死区,在梗死区域内分化为类心肌样细胞,穴位注射与静脉注射比较无明显差异。

Abstract:Objective:To discuss the situation of myocardial cells freshman differentiation in the infarcted after myocardial infarction by points and the vein graft in 0 days and 7 days.Methods:With anterior descending coronary artery ligated way model of acute myocardial infarction,models of myocardial infarction in Japan white rabbits were randomly divided into 5 groups,Japan’s big ears white rabbit from healthy bone marrow,separate training of bone marrow stromal stem cells and ChuanDai,In acute myocardial infarction after forming 0 days and 7 days acupuncture points and intravenous stem cells.Brdu immunohistochemical staining,take a slice of brdu positive cells to immunohistochemical stains,a microscope myocardial specificity ctni dyeing.Results:Model control group myocardial infarction area did not see the brdu markers of bone marrow stromal stem cells;Point injection stem cells 0 days brdu group of positive cells average light density values more intravenous stem cells 0 days increase trend of obviousP<0.05;Point injection stem cells 7 days brdu group of positive cells average light density values more intravenous stem cells 7 days increased group no obvious differenceP>0.05.Conclusion:In 0 days meridians and intravenous bone marrow stromal stem cell migration to infarction area,in the infarction area live and differentiation is kind of myocardial appearance cell;In 7 days meridians and intravenous bone marrow stromal stem cell migration to infarction area,in the infarction area for the kind of myocardial appearance cell differentiation,point injection and intravenous there were no significant differences.
