松龄血脉康胶囊对自发性高血压大鼠肝阳上亢证相关指标的影响

目的:研究松龄血脉康胶囊对自发性高血压大鼠肝阳上亢证的血压影响以及调节机制。方法:24只10周龄自发性高血压大鼠(SHR)随机分为对照组、模型组和中药组,每组8只。第1周,模型组和中药组灌附子汤,20mL/(kg.d)灌胃,相当生药2g/(kg.d);第2～5周,中药组灌松龄血脉康胶囊20mg/(kg.d),对照组和模型组灌同体积的生理盐水。每周观察大鼠的外观体征和行为活动等变化,观察大鼠易激惹程度并分级,每周测血压和称体重1次,第5周检测各组大鼠痛阈和脑组织五羟色胺(5-HT)mRNA的表达量。结果:松龄血脉康给药4周后,可有效抑制血压上升,调控血压使血压有逐渐下降的趋势;改善大鼠急躁易怒的症状;显著上调脑组织5-HT mRNA的表达。结论:上调脑组织5-HT的表达,可能是松龄血脉康胶囊治疗肝阳上亢型高血压的机制之一。

Effects of Songling Xuemaikang Capsule for Spontaneously Hypertensive Rats with Hyperactivity of Liver Yang

Objective:The objective of this study was to research the blood pressure adjustment mechanism of Songling Xuemaikang Capsule treated spontaneously hypertensive rats with hyperactivity of liver yang.Methods:24 10-week-old spontaneously hypertensive rats(SHR)were randomLy divided into 3 groups including control group,model group and Chinese medicine group.There were 8 rats in each group.In the 1st week,rats in model group and Chinese medicine group were fed with Aconite Decoction 20mL/(kg·d)(intragastric administration).The medicine was fairly crude drugs 2g/(kg o d).From 2nd to 5th week,Songling Xuemaikang Capsule 20mg /(kg o d)was given to Chinese medicine group,while the same volume of saline was fed to control and model groups.The appearance of signs and behavior changes,degree of rats′ irritability,blood pressure and weight were observed once a week.The pain threshold and the expression of 5-HT mRNA in brain tissue were measured in the 5th week.Results:The increased blood pressure could be inhibited and declined gradually after 4 weeks′ treatment of Songling Xuemaikang Capsule.It could also improve the irritability symptoms of rats.5-HT mRNA expression was significantly raised in brain tissue.Conclusion:Raising expression of 5-HT in brain tissue may be one of the mechanisms in Songling Xuemaikang Capsule for hyperactivity of liver-yang hypertension.
