根管桩用石英纤维复合材料及其树脂基体的细胞毒性研究

摘    要:目的:研究白行研制的根管桩用复合材料及其环氧树脂基体的体外细胞毒性,以初步评价材料的生物安全性。方法:采用四唑盐比色法(MTT法),分别用50%和100%的石英纤维复合材料以及50%和100%环氧树脂的浸提液培养小鼠成纤维细胞L-929细胞2d、4d、7d后,测定吸光度值(OD值),并计算细胞相对增殖率,以6级毒性分类法评级,采用单因素方差分析法对各实验组及对照组的OD值进行统计学分析,显微镜观察细胞的生长状况。结果:各观察期细胞生长良好,形态正常,复合材料及其环氧树脂基体的细胞毒级为0—1级。结论:自行研制的根管桩用复合材料及其树脂基体无细胞毒性。

Abstract:Objective: To investigate the cytotoxicity of custom-made fiber-reinforced composite and its epoxy resin matrix utilized for root canal post, in order to evaluate the biocompatibility of materials primarily. Methods:Colorimetric MTT(tetrazolium)assay was used, mouse lung fibroblast L-929 were cultured in the 50%?100% extraction of composite and its epoxy resin matrix for 2d, 4d, 7d respectively. Light absorption values(OD) were measured with enzyme-linked immunoassay instrument, cell relative growth rate(RGR)were calculated. Cytotoxicity was evaluated with 6 grade classification and OD values were analyzed statistically by ANOVA. Cell growth status was observed under microscopy. Results: L-929 cells showed normal proliferation and morphology during observation periods, the cytotoxicity of custom-made fiber-reinforced composite and its epoxy resin matrix was 0-1grade.
