缺氧诱导因子-1α在胃腺癌中的表达及其与肿瘤细胞增殖、凋亡的关系

摘    要:目的 探讨缺氧诱导因子(HIF)-1α在胃腺癌中的表达及其对肿瘤细胞增殖、凋亡的影响和临床意义.方法 采用免疫组织化学SP法检测100例胃腺癌组织中HIF-1α、Cyclin D1和bcl-2的表达.结果 随着胃腺癌分化程度的降低、浸润深度的增加及发生淋巴结转移,HIF-1α、Cycfin D1阳性表达率也增加(X2值分别为4.99、7.20、11.65、5.36、4.55、4.58;P<0.05或P<0.01);随着浸润深度的增加及发生淋巴结转移,bcl-2的表达也明显升高(值分别为6.21、7.66;P<0.05或P<0.01).胃腺癌中Cyclin D1与bcl-2表达呈显著正相关(,值为5.96,P<0.05).HIF一1α阳性表达者中Cyclin D1、bel-2的阳性表达率均明显高于其阴性表达者,HIF-1α表达与CyclinD1、bcl-2的表达明显相关(X2值分别为5.94、6.14;P均<0.05).结论 三者的检测对判断胃腺癌的浸润转移能力及预后具有重要的临床意义;胃腺癌的浸润转移在一定程度上是肿瘤细胞不断增殖和凋亡抑制的结果 ;HIF-1α在一定程度上能够诱导肿瘤细胞的增殖、抑制其凋亡而促进胃腺癌的发生发展.

Effect of hypoxia-inducible faetor-1α expression on proliferation and apoptosis of gastric adenocar-cinoma
Abstract:Objective To explore the effect of hypoxia-indueible faetor-1α expression on proliferation and apoptosis of primary gastrie adenoeareinoma cells and its elinieal significance. Methods The expression of HIF-1α,Cyelin D1 and bel-2 was detected by using SP immunohistoehemieal staining in 100 eases of gastric adenoeareinoma tissues. Results The expression of HIF-1α and Cyelin D1 was correlated with tumor differentiation, invasive depth and metastasis of tumors (X2=4.99,7.20,11.65,5.36,4.55,4.58 ,P <0.05 or P <0.01 ). The expression of bd-2 was correlated with invasive depth and metastasis (X2 = 6.21,7.66, P < 0.05 or P < 0.01 ). There was a strong association between Cyelin D1 and bel-2 expression (X2= 5.96 ,P<0.05 ). There was also a strong association between HIF-1α expression and Cyc-lin D1 or bel-2 (X2=5.94,6.14,P<0.05). Conclusion Detection of HIF-1α,Cyelin DI and bel-2 expression may be helpful to evaluate invasion and metastasis and prognosis of gastric adenoeareinoma. HIF-let can induce the proliferation of tumor cells and suppress their apoptosis to some extent,thereby promoting the development of gastric adenoearcinoma.
