低频超声辐照兔胚胎后胎盘组织细胞p53、Bcl-2的表达改变

摘    要:目的探讨超声止孕的可能性。方法低频聚焦超声(353kHz)6种声功率直接辐照妊娠第10d兔胚胎,10d后观察胚胎死亡率,并用免疫组化和原位杂交法检测胎盘组织细胞p53、Bcl-2的变化。结果声功率30W/cm^2、35W/cm^2及≥40W/cm^2组的胚胎死亡率分别为25.00%、72.88%和100%;辐照后胎盘组织细胞p53表达增加,96h升至最高点;Bcl-2表达下降,96h下降至最低点,以后两者均有所恢复。结论低频超声抗孕具有潜在的可行性,可诱导胎盘组织细胞p53表达增加,Bcl-2表达下降,但具有可复性。

Abstract:Objective To investigate the availability of early pregnancy termination with low frequency ultrasound. Methods The 10th day rabbit embryos were irradiated with low frequency focused ultrasound. Embryonic mortalities and expressions of p53 and Bcl 2 detected by immunohistochemistry and in situ hybridization were observed 10 days after irradiating. Results The embryonic mortalities were 25.00% ,72.88% and 100% respectively with an intensity of 30 W/cm^2,35 W/cm^2 and equal or more than 40 W/cm^2 for 60 seconds. The incidence of p53 expression of placenta cell was increased after irradiating, peaked at 96 hours,and the expression of Bcl-2 was decreased at 24 hours and least at 96 hours after irradiating. Then both level recovered slowly. Conclusions It is feasible for early pregnancy termination with low frequency ultrasound. The low frequency ultrasound may result in more p53 and less Bcl-2 expression of placenta cells, but both can be resumptive.
