摘    要:目的研究大肠癌组织中多肿瘤抑制蛋白(p16)、细胞周期蛋白(cyclinD1)及细胞周期蛋白依赖激酶(CDK4)的表达及其意义。方法采用组织芯片技术制作80例大肠癌组织芯片,同时用S-P免疫组织化学方法检测大肠癌组织芯片中p16、cyclinD1和CDK4的表达。结果80例大肠癌中p16、cyclinD1和CDK4的阳性率分别为40.0%、57.5%、47.5%。不同组织学类型、淋巴结转移状况的大肠癌中,p16、cyclinD1的表达无明显差异(P>0.05)。不同组织学类型、浸润程度及淋巴结转移状况的大肠癌中CDK4的表达均无明显差异(P>0.05)。结论p16表达、cyclinD1高表达与大肠癌的浸润程度密切相关,大肠癌中p16低表达与cyclinD1的高表达密切相关。p16低表达影响cyclinD1的高表达。应用组织芯片大规模高效检测临床组织样本是可行的,具有快速、方便经济、准确的特点。

Abstract:Objective To study the expressions of p16,cyclinD1,CDK4and anal-yse their significances in large intestinal cancer.Methods Using tissue chip technique to prepare for tissue chips for80cases with large intestinal cancer,while using the S-P immunohistochemical method.The expressions of p16,cyclinD1and CDK4in large in-testinal cancer tissue chips were studied.Results The positive rates of p16,cyclinD1and CDK4were40.0%,57.5%and47.5%in80cases with large intestinal cancer re-spectively.No difference was found between the expressions of p16and cyclinD1in dif-ferent tissue types and metastasis in lymph node originated large intestinal cancers(P>0.05).A significant difference was found between the expressions of p16and cyclinD1in different invasion of large intestinal cancers(P<0.05).No difference was found be-tween the expressions of CDK4in different types,metastasis in lymph node,and inva-sion degree in large intestinal cancer(P>0.05).Conclusions The low expression of p16,overexpression of cyclinD1and invasion of large intestinal cancer have significant corre-lation.It is a significant correlation between the low expression of p16and overexpres-sion of cyclinD1in large intestinal cancers.The low expression of p16has a role to ef-fect the overexpression of cyclinD1in large intestinal cancer.It is practical to utilize tissue chip which is a rapid,economic and accurate to screen the clinical tissue speci-mens on a large scale.
