牙髓干细胞体外三维培养及生物学特性的研究

摘    要:目的建立大鼠牙髓干细胞(DPSC)体外培养体系及分析研究细胞因子骨形成蛋白(BMP-2)对其生物学特性的影响。方法利用一步离心法形成细胞团块或聚集物建立DISC三维球状培养体系,观察细胞在培养过程中形态和细胞基质合成的变化,并分析研究细胞因子BMP-2对于三维培养DPSC增殖和分化的影响。结果三维培养的DPSC数目无明显增加,细胞基质沉积,ALP的含量增加,活性增高;在三维培养体系中,BMP能够增加ALP含量,且呈浓度依赖性。结论三维培养体系中形成的细胞外基质作为天然的支架,由BMP-2持续释放和保留,对今后的龋齿和牙髓治疗的临床应用有一定价值。

Three-dimension culture and biological characteristics of dental pulp stem cell

Abstract:Objective To establish the model of 3-dimension culture model of rat DPSCs and to investigate the effects of BMP-2 on the biological characteristics of the model.Methods Three-dimension globular culture model was established by cell aggregate with one step centrifugalization. Cell shape and cell matrix were observed during culture procedure. The effects of BMP-2 on the proliferation and differentiation of DPSCs in the 3-dimension culture were analyzed.Results The number of DPSCs in the 3-dimension culture did not increase. It was found that cell matrix deposited. The content of ALP and the activity of ALP were both increased. BMP increased the content of ALP in a dose-dependent manner.Conclusion The natural supporter, which is formed by the extracellular matrix in the 3-dimension culture model with facilitation of BMP, will be valuable for the clinical application to pulp disease.
