摘    要:目的建立高效液相色谱法测定尿毒清颗粒中大黄酸含量的方法。方法采用HPLC法,色谱柱:Phenomenex(250 mm×4.6 mm,5μm),流动相:甲醇-0.1%磷酸溶液(85∶15),柱温:室温,流速:1.0 ml.min-1,检测波长:254 nm。结果大黄酸在0.834 5μg.ml-1~25.035μg.ml-1内呈良好线性,R2=0.999 6,平均回收率为100.34%,RSD为1.06%(n=9)。结论本方法准确、精密度高、专属性好,可用于尿毒清颗粒的质量控制。

Abstract:Objective To determine the contents of Rhein in Niaoduqing grains by means of the HPLC(High Performance Liquid Chromatography).Methods A HPLC has been established by the mobile phase of methanol-0.1% H_3PO_4 aqueous solution for Rhein in Niaoduqing grains,using a Phenomenex column(250 mm×4.6 mm,5 μm).Detect wavelength was 254 nm,column temperature was room temperature,flow rate was 1 ml·min-1 and injection volume was 10 μl.Results The celibration curve was linear in the range of 0.834 5 μg·ml-1~25.035 μg·ml-1 for Rhein,with R2 of 0.999 6,the average recovery of 100.34% with the RSD of 1.06%.Conclusion The method is reliable,accurate and specific,which can be used for the quality control of Niaoduqing grains.
