脊髓损伤早期一氧化氮代谢变化及其对脊髓神经功能影响的实验研究

摘    要:目的探讨脊髓损伤早期NO的代谢变化情况及其与脊髓神经功能的关系。方法以大白鼠为实验动物,用Alien’s打击器打击骨髓,建立中度及重度脊髓损伤模型,测量脊髓损伤后4h、12h、24h、3d、1周、2周脊髓损伤局部及血清中NO含量,并观察NO含量变化与脊髓运动神经功能恢复情况之间的关系。结果脊髓损伤后血清NO含量无明显变化,而损伤脊髓局部的NO含量在伤后4h已明显增高,在24h后达到高峰并持续2周以上,在中度脊髓损伤情况下,脊髓运动神经功能自然逐步恢复,在重度脊髓损伤情况下,脊髓运动神经功能不能恢复,而这两种脊髓损伤情况下,伤后早期损伤局部的NO含量变化情况相似。结论脊髓损伤后,NO的代谢变化表现为损伤局部NO含量迅速升高,伤后早期损伤局部NO含量迅速升高不是脊髓继发损伤的原因。

Abstract:Objective Exploration of relationship between metabolic change of NO in early period afterspinal cord injury and recovery of motor neurologic deficit of spinal cord Methods With rat asexperimental animsal, moderate and severe injury of spinal cord were established. The concentration of NOin ijuried spinal cord and serum was measured in 4h, 12h,24h, 3days, 1 week and 2weekes after spinal cordinjury. The relationsip between the change of concentration of NO and the enery of motr neurologicdeficit of spinal cord was observed Results After spinal cord injury, the concentration of NO in serumdid not alter. The colcentation of ON in injdried spinal cord ed toificantly in 4h after spinal cordinjury, reaching to peak in 24h and persisting more than two weekes. The motor neurologic deficit ofspinal cord recoveried gradualy when the prirnary injury of spinal cord was moderate and did not improvewhen the primary injury of spinal cord was serious. In these two cases, the change of quantity of NO ininjured spinal cord were similar. Conclusions In eary period after spinal cord injury, the metabolicchange of NO should be that the concentration of NO in injuried spinal cord increase quickly. This changeshould not result in secondary injury of spinal cord.
