摘    要:目的研究去甲二氢愈创木酸(NDGA)对小鼠胚泡植入的影响及其作用机制。方法孕2d小鼠随机分4组(A、B、C和对照组),A、B、C组分别经皮下注射1.0、3.0、9.0mg/kg的NDGA,对照组注射等量PBS溶液,孕4d处死一半,取子宫免疫组化检测基质金属蛋白酶-9(MMP-9)表达,透射电镜分析子宫内膜结构的变化;另一半于检孕8d处死,测子宫脏器系数,观察胚胎着床数。结果与对照组比较:A组子宫脏器系数和妊娠胚胎数无显著差异,B、C组子宫脏器系数显著下降,妊娠胚胎数明显减少;A组子宫内膜MMP-9阳性表达差异无显著性,而B、C组MMP-9阳性表达明显减少;A组子宫内膜细胞超微结构无显著变化,B、C组细胞超微结构均遭受不同程度的损伤。结论NDGA可能通过对子宫内膜细胞直接毒性作用干扰MMP-9的表达,抑制胚泡植入。

Abstract:Objective To investigate the effect of nordihydroguaiaretic acid (NDGA) on blastocyst implantation and its mechanism in mice. Methods Pregnant mice was randomly divided into four groups(A,B,C and control).A, B and C groups were given NDGA(1.0, 3.0, 9.0 mg/kg)by sc injection on the embryonic day 2 (E2) and the control group were given the equal volume of PBS. Half of mice were killed on E4 to observe the immunohistochemical and ultrastructural variances in endometrium, the other half were killed on E8 to examine the number of the embryo and calculate the uterine organ coefficient. Results There were no significant diffrences in the number of the embryo and the uterine organ coefficient between A group and control. Compared with the control, the number of the embryo and the uterine organ coefficient were decreased in the B group; No embryo was observed in C group. Compared with the control the expression of MMP-9 in the endometrium was decreased in the B and C group, but there were no significant diffrences between A group and control.Compared with the control, the ultrastructure of decidual cells and glands in endometrium in the B and C group were damaged, but there were no significant diffrences in the damage of the endometrium between A group and control. Conclusion NDGA may be via destroying the endometrial cell, affecting expression of MMP-9 inhibits the implantation of embryo in the early pregnancy.
