兔颅脑创伤后血清抗脑抗体变化及脑组织谷氨酸的表达

摘    要:目的:研究家兔颅脑创伤后抗脑抗体的变化及脑组织谷氨酸的表达。方法:16只成年新西兰白兔分为两组,颅脑创伤组(n=8)、假手术对照组(n=8),建立兔脑单侧重型液压冲击创伤模型,收集所有家兔创伤前2h及创伤后3、7、14、21、28、35、42、49d各时间点血清标本。用ELISA法检测血清抗脑抗体的浓度:用免疫组化方法检测脑组织谷氨酸(Glu)的表达。结果:在予以单侧液压打击后,颅脑创伤组家兔血清中抗脑抗体浓度增高并有峰值出现,假手术对照组抗脑抗体浓度变化则不明显,两组比较差异有统计学意义(P〈0.05);颅脑创伤组家兔脑组织Glu的表达较假手术组增高,两组比较差异有统计学意义(P〈0.05)。结论:颅脑创伤后,血清中抗脑抗体与伤前相比有明显升高;颅脑创伤后脑组织Glu的表达增高,提示抗脑抗体可能是颅脑创伤后出现癫痫的原因之一。

Abstract:Objective: To study the changes of the antibrain-antibody (ABAb) in blood serum and expression of Glu in cerebral cortex after the traumatic brain injury. Methods: The rabbits were divided into TBI group (n=8) and sham operation group (n=8). An animal model of severe lateral fluid percussion brain injury(LFPI) were established. All group rabbits were phlebotomized blood 4 ml through the ear fringe vein at 2 h before injury, 3,7,14,21,28,35,42,49 d after injury. Antibrain-antibody was determined by ELISA, and expression of Glu was examined by immunohistochemistry. Results: After LFPI,TBI group had the peak value of the ABAb, while the sham operation group had no significant change. There was statistically significant difference between TBI and sham operation group (P 〈 0.05). After brain trauma, the Glu expression levels were obvioursly higher than those in the sham operation group(P 〈 0.05). Conclusion: After trauma brain injure,the concentration of ABAb is higher than nature. After brain trauma, the concentration of ABAb and the Glu expression level is higher than nature, the increased ABAb will be likely to attack self brain tissue and induce epilepsy.
