为探讨中国人北方汉族２型ＤＭ醛糖还原酶（ＡＲ）基因５’端的（ＡＣ）ｎ的多态性与糖尿病视网膜病变易感性的关系，用＾３２ＰｄＣＴＰ掺入ＰＣＲ，６％甲酰胺聚丙烯酰胺凝胶电泳和放射性自显影的方法，分析了１１６例２型ＤＭ「包括无并发症（ＮＤＣ）组６６例，视网膜病变（ＤＲＰ）组５０例」和４２例正常对照组（ＣＯＮ）ＡＲ基因５’端（ＡＣ）ｎ的多态性。结果显示，ＤＭ组和对照组均发现７种等位基因Ｚ－６～Ｚ＋６，Ｚ－２

STUDY ON THE RELATIONSHIP BETWEEN THE POLYMORPHISM OF (AC)n IN THE 5'-END OF THE AR GENE AND THE SUSCEPTIBILITY OF MICROANGOPATHY IN TYPE 2 DIABETES MELLITUS

To study the relationship between the polymorphism of (AC)n in 5'-end of the AR gene and the susceptibility of DMIP in type 2 DM of Han nationality in North China, 116 patients with type 2 DM(66 patients had no DMIP(NDC) and 50 patients with diabetic retinopathy(DRP))and 42 healthy persons as control (CON)group were included. Genomic DNA was prepared from white blood cells. PCR was performed after mixed into 32 PdCTP. PCR products were electrophoresed with 6% formamide/urea gel and autoradiogaphy was performed. Results showed 7 types of alleles (Z-6~Z 6). The frequency of allele Z-2 was significently higher in DRP group than in NDC group and CON group, but the the frequency of allele Z 2 was much lower. In type 2 DM of Han nationality in North China, AR gene (AC)n presents 7 alleles. Z-2 allele might be the susceptible gene and Z 2 allele might be the protected gene.
