黄嘌呤抗视神经切断后视网膜细胞凋亡的作用

摘    要:目的 探讨眼球玻璃体注射 3 异丁基 1 甲基 (IBMX)对切断视神经后视网膜细胞凋亡的作用。方法 用荧光核染料 332 5 8和琼脂糖凝胶电脉技术观察切断视神经 5、7和 1 4d后成年金黄地鼠视网膜细胞凋亡的变化。结果  ( 1 )视神经切断 5、7、1 4d后 ,节细胞层 (GCL)细胞凋亡密度 (cell/mm2 )分别为 94 60± 2 9 40、81 2 0± 1 5 1 9和 31 2 0± 1 1 43,给予IBMX组在上述时间点凋亡密度为 67 80± 1 3 83、60 2 0± 9 68和 2 7 0 0± 9 5 7。IBMX 7d组与对照组比较有显著差异 (P <0 0 5 )。 ( 2 )琼脂糖凝胶电脉显示切断 7d的视网膜细胞DNA抽提物显示典型的DNA梯形条带。其它各间间点 ( 5d和 1 4d)以及玻璃体内注射IBMX均未见典型DNA梯形条带。结论 IBMX有抗视神经切断后视网膜细胞凋亡的作用

Abstract:Objective To investigate the effects of 3 isobutyl methylxanthine(IBMX) on apoptosis of retinal cells after optic nerve transection.Methods Hoechst 33258 fluorescent staining and detection of DNA fragmentation by agrose gel electrophoresis was used to observe the change of apoptotic cells in retina in all groups of adult hamsters.Results (1)At 5?7?14day after optic nerve transection(ONTS group),the mean densities of apoptotic cells in the layer of ganglion cell(GCL)are 94 60±29 40?81 20±15 19 and 31 20±11 43.In the group treated with IBMX(IBMX group),the mean denstities are 67 80±13 83?60 20±9 68 and 27 00±9 57,respectively.There are significant difference ONTS group compared with IBMX group at 7 day, P<0 05.(2)The result of DNA electrophoresis indicated that only the extraction of retinas which transected for 7 days show typical DNA ladder,while the other groups(5,14day and IBMX groups)did not show typical DNA ladder.Conclusions IBMX may have the capacity of preventing apoptisis.
