迪康凝胶对大鼠烫伤创面的治疗作用及可能机制研究

目的 探讨迪康凝胶对烫伤大鼠创面愈合的效果及作用机制.方法 将SD大鼠随机分为模型组、美宝烫伤膏组、迪康凝胶组,3组均采用超级控温烫伤仪,造背部深Ⅱ度烫伤.模型组不给药,其余给药1次/d,连续21 d.连续考察21 d内创面愈合情况及创面病理组织形态.ELISA法检测组织TNF-α和IL-8及Ⅰ 、Ⅲ型胶原.RT-PCR检测组织TNF-α和IL-8表达水平.结果 与模型组比较,迪康凝胶组愈合时间缩短(P<0.05,P<0.01),创伤愈合率增高(P<0.05,P<0.01),差异有统计学意义.组织学观察证明,迪康凝胶具有良好的组织损伤修复作用.ELISA检测表明,烫伤后两药物组创面组织TNF-α和IL-8含量均明显低于模型组(P<0.05,P<0.01);而胶原Ⅰ 、Ⅲ组织含量均高于模型组.RT-PCR检测表明,两药物组组织TNF-αmRNA和IL-8 mRNA表达水平均与其蛋白浓度水平基本一致.结论 迪康凝胶能促进大鼠皮肤烫伤后创面愈合,是治疗烫伤创面的有效药物.

A Study on the Therapeutic Effect and Probable Mechanisms of Dikang Gel on the Scald Wound Healing of Rats

Objective To investigate the therapeutic effect and mechanism of Dikang Gel on the scald wound healing of rats .Methods The SD rats were divided randomly into the model group ,Mebo Ointment group ,and Dikang Gel group .The deep second-degree scald model was established with a super temperature-controlled scald apparatus in SD rats .The model group was not treated ,while the other two groups were administered once a day for 21 days and the wound healing process and histopathology were monitored within 21 days .The levels of TNF-α, IL-8 ,typeⅠand Ⅲ collagen in tissues were detected by the ELISA method and the mRNA expression of TNF-αand IL-8 in tissues were detected by RT-PCR .Results The healing time was significantly shorter (P<0.05 ,P<0.01) and the wound healing rate was significantly higher (P<0.05 ,P<0.01) in the Dikang Gel group than in the model group .The results of histological observation showed that Dikang Gel had a good effect of tissue repair .The levels of TNF-αand IL-8 in the wound tissues were significantly lower in the two administered groups than in the model group (P<0.05 ,P<0.01) .The levels of typeⅠand Ⅲ collagen were also higher in the two administered groups than in the model group .The results of RT-PCR showed that the mRNA expression levels of TNF-αand IL-8 in the two administered groups were consistent with the levels of protein concentration .Conclusion Dikang Gel can promote the scald wound healing of rats ,so it is an effective drug in the treatment of the scald wound .
