麦芽醇对大鼠局灶性脑缺血再灌注损伤后神经元凋亡的影响

摘    要:目的观察麦芽醇对大鼠局灶性脑缺血再灌注损伤后海马神经元内Caspase-3和神经元凋亡的影响。方法大脑中动脉线栓法建立脑缺血再灌注损伤模型,应用免疫印迹法和流式细胞计数法分别检测Caspase-3和细胞凋亡。结果再灌注72h后海马Caspase-3表达升高,凋亡细胞增多,麦芽醇组Caspase-3表达和凋亡细胞明显减少。结论麦芽醇对脑缺血再灌注损伤有保护作用。

Abstract:Objective To investigate the effects of maltol on expression of Caspase-3 protein in neurons and neuronal apoptosis after focal cerebral ischemia-reperfusion in rats.Methods A model of middle cerebral artery occlusion(MCAO)in rats was established with intraluminal filament occlusion.The Western-blot and flowcytomete were used respectively to observe the quantities of Caspase-3 protein and neuronal apoptosis in hippocampus.Results A significant increase in Caspase-3 immunoreactivity and apoptosis were detected at 72h after reperfusion.The difference of Caspase-3 and apoptosis between ischemic group and maltol group were significant.Conclusion Maltol can protect cerebral tissues by inhibiting cell apoptosis in ischemia reperfusin damages.
