1型糖尿病患者血浆血小板活化因子乙酰水解酶(PAF-AH)活性检测及其基因单核苷酸多态性的研究

摘    要:目的观察1型糖尿病(T1DM)病人血浆血小板活化因子乙酰水解酶(PAF—AH)的活性以及PAF.AH基因G275A多态性与1型糖尿病(T1DM)的相关性。方法选择T1DM病人115例,对照组106例,测定血浆PAF-AH活性,采用PCR-RLFP法测定PAF-AH基因G275A基因型。结果T1DM患者的血浆PAF.AH活性为(18.95±5.43)nmoL/(min·ml);健康对照者血浆PAF-AH活性为(26.65±3.71)nmol/(min·ml),2组比较差异具有统计学意义(P〈0.05)。与对照组相比,TIDM组G275A多态性位点GA基因型频率显著高于对照组(36.52%VS22.64%,P〈0.05)。结论T1DM患者的血浆PAF-AH活性降低。PAF-AH基因G275A多态性可能与1型糖尿病相关。

Abstract:Objective To investigate the plasma PAF-AH level in type 1 diabetes patients and the association between the PAF-AH gene polymorphism and type 1 diabetes mellitus. Methods Plasma PAF-AH in115 type 1 diabetes patients and 106 controls were determined by PAF-AH assay kit, and PAF-AH gene G275A genotypes were detected by PCR-RLFP. Results The activity of plasma PAF-AH in the patients with type 1 diabetes was significantly lower than that in the controls. Compared with control group, the frequencies of the GA genotype on G275A polymorphism were significantly higher than that in T1DM group. Conclusion The activity of plasma PAF-AH in the patients with type 1 diabetes was significantly lower than that in normal controls. The G275A polymorphism of PAF-AH gene might be related to type 1 diabetes mellitus.
