摘    要:目的探讨高血糖对脑缺血后血脑屏障(BBB)葡萄糖转运蛋白1(GLUTl)表达的影响。方法采用链脲佐菌素法建立大鼠高血糖模型,线栓法制作大鼠大脑中动脉闭塞模型,用免疫组化法观察脑缺血后1h、3h、6h、12h、1d、2d、3d、7d时BBBGLUT1表达的变化。结果正常血糖组大鼠及高血糖组大鼠脑缺血后BBBGLUT1的表达均增加,除脑缺血后7d外,高血糖组脑缺血后各时间点GLUT1的表达均低于正常血糖组(P〈0.01)。结论高血糖可降低脑缺血后BBBGLUT1的表达,隆低BRB蒴菊搐接请活性

Abstract:Objective To investigate the influence of hyperglycemia on the expression of glucose transporter1(GLUT1) in blood-brain barrier(BBB) after cerebral ischemia in rats.Methods We used streptozotocin(STZ) to induce diabetic experimental models and made focal permanent cerebral ischemia models by occluding the middle cerebral artery with nylon line in rats.The changes of GLUT1 expression in BBB were investigated by immunohistochemistry staining.Results It was found that the expression of GLUT1 in BBB increased after cerebral ischemia.It was much lower than that of normoglycemic rats at every time point except 7d after cerebral ischemia(P<0.01).Conclusions Hyperglycemia has the effect that it can reduce the expression of GLUT1 in BBB following cerebral ischemia,as well as the transportation of glucose in BBBB.
