复方有效部位与组方药材指纹图谱色谱峰相关性研究

摘    要: 目的在色谱指纹图谱分析的基础上,研究复方有效部位与组方药材指纹图谱色谱峰相关性,进而分析有效部位化学组成。方法以精制血府逐瘀汤降血脂有效部位(FP)为研究对象,通过HPLC/DAD联用建立复方有效部位及各药材相应部位指纹图谱,以色谱峰保留时间相对偏差△t和紫外光谱相似度为考察指标,对方剂有效部位化学指纹图谱色谱峰归属进行分析。结果对复方有效部位FP中的16个色谱峰进行了归属分析,确定其主要来源于枳壳、川芎、赤芍、红花,为黄酮及萜类化合物。结论该方法能较好地判别FP部位主要色谱峰与药材化学组分间的相关性,帮助确定FP部位指纹峰的归属,可应用于中药复方有效部位化学组成及来源分析。

Abstract:OBJECTIVE Based on the chromatographic fingerprint analysis of traditional Chinese medicine(TCM), a new method for correlativity analysis between fingerprint peaks and the effective fraction (FP) of TCM was proposed.METHODS Using the effective fraction FP of Xuefuzhuyu decoction as an example,the chromatographic fingerprints of FP and the relevant fractions of its herbs were configured by HPLC/DAD analysis.The relative deviation △t of retention time and the similarity of UV spectra of peaks were utilized as indices to evaluate the correlativity.RESULTS The assignment results of 16 peaks of FP fraction indicated that the components of FP were originated primarily from Fructus aurantii,Ligusticum chuanxiong Hort,Radices paeoniae rubra and Carthamus tinctorius,which belonged to flavonoids and terpenes.CONCLUSION The established method effectively establised the correlativity between FP and its relevant fractions, which would contribute to deduce the chemical components or sources of effective fraction of TCM.
