急性肺血栓栓塞症时血红素氧合酶-1 mRNA的表达

目的 观察急性肺血栓栓塞症时,血红素氧合酶-1(HO-1)mRNA的表达及变化.方法 健康成年雄性杂种犬24只,随机分为4组:对照组、肺栓塞3 h组、肺栓塞8 h组、肺栓塞24 h组.经右颈静脉置管注入自体血栓,股静脉置入Swan-Ganz监测血液动力学,采用半定量RT-PCR技术测定急性肺血栓栓塞症不同时相HO-1 mRNA的相对表达量.结果 急性肺血栓栓塞症3 h可见有HO-1 mRNA的表达并逐渐升高;犬肺动脉压在肺栓塞即刻显著升高,之后呈下降趋势.结论 急性栓塞性肺动脉高压HO-1 mRNA表达增多,HO-1 mRNA表达与肺动脉压呈负相关性.

Study on changes of hemo oxygenase-1 mRNA expression in acute pulmonary thromboembolism in dogs

Objective To investigate the changes of hemo oxygenase-1(HO-1) mRNA expression in acute pulmonary thromboembolism in dogs.Methods 24 healthy grown male dogs were divided randomly into 4 groups: control group,pulmonary embolism 3 h group,pulmonary embolism 8 h group and pulmonary embolism 24 h group.Catheter was placed by the right jugular vein to inject the auto-thrombus.Swan-Ganz catheter was placed by the femoral vein to observe the changes of hemodynamics.The quantity of hemo oxygenase-1 mRNA expression in acute pulmonary thromboembolism was detected by semi-quantity RT-PCR method.Results The expression of hemo oxygenase-1 mRNA in acute pulmonary thromboembolism increased gradually post 3 h the pulmonary embolism.The pulmonary arterial pressure increased remarkably at the time of embolized,and decreased gradually after the pulmonary embolism.Conclusion The expression of HO-1 mRNA increased in acute pulmonary thromboembolism.And it has a negative relativity with the pulmonary arterial pressure.
