槲皮素对宫颈癌细胞SiHa增殖的影响

摘    要:目的:探讨槲皮素对于人宫颈癌细胞Si Ha的作用及机制。方法:用不同浓度的槲皮素(0、20、40和80μmol/L)处理Si Ha细胞不同时间后(24 h,48 h和72 h),通过噻唑蓝(MTT)法检测细胞活力的变化;通过流式细胞仪检测槲皮素对细胞周期的影响;通过逆转录聚合酶链反应(RT-PCR)和蛋白质印迹法(Western blotting)检测槲皮素对细胞周期蛋白D1(Cyclin D1)表达的影响。结果:MTT法检测结果显示,槲皮素处理Si Ha细胞24 h和48 h后,40μmol/L和80μmol/L组的细胞活力低于对照组(P0.05);槲皮素处理Si Ha细胞72 h后,20μmol/L、40μmol/L和80μmol/L组的细胞活力低于对照组(P0.05);在40μmol/L和80μmol/L组,随着培养时间延长,细胞活力较24 h时降低(P0.05)。流式细胞仪检测结果显示,Si Ha细胞经过20、40和80μmol/L浓度的槲皮素分别处理48 h后,处于G0/G1期的细胞百分比明显高于对照组。RT-PCR和Western blotting结果显示,槲皮素可以下调Si Ha细胞中Cyclin D1蛋白的表达。结论:槲皮素通过下调Si Ha细胞中Cyclin D1蛋白的表达,可以抑制Si Ha细胞的活力和增殖。

Abstract:Objective: To investigate the effect and mechanism of quercetin on human cervical cancer cell line SiHa. Methods:MTT assay was used to detect the effect of quercetin on proliferation of cervical cancer cell SiHa at different times (24 h, 48 h and 72 h) and different concentrations (0, 20, 40 and 80μmol/L);flow cytometry was used to detect the cell cycle in quercetin treated SiHa cells;RT-PCR and Western blotting was used to detect the expression of Cyclin D1 in quercetin treated SiHa cells. Results:The cell viability of SiHa cells by treating with quercetin for 24 h and 48 h in 40 μmol/L and 80μmol/L group were significantly lower than control group;The cell viability of SiHa cells by treating with quercetin for 72 h in 20μmol/L, 40μmol/L和80μmol/L group were significantly lower than control group;the number of G0/G1 cells in 20μmol/L, 40μmol/L and 80μmol/L group was significantly more than control group by treating with quercetin for 48 h;the protein level of Cyclin D1 was down-regulated in quercetin treated group. Conclusions: Quercetin could inhibit cell vitality and cell proliferation by down-regulating the expression of Cyclin D1 in cervical cancer cell SiHa.
