摘 要: | 目的 :研究NFkB家族蛋白在乳腺癌细胞的表达及与乳腺癌细胞化疗耐药性关系。方法 :培养 14个乳腺癌细胞系 ,对其进行胞浆、胞核蛋白质提取 ,Western印迹法观察NFKB家族蛋白及其抑制物的表达 ,并用MTT药敏实验观察NFkB家族蛋白与乳腺癌细胞化疗耐药性关系。结果 :p5 0、p65和IkB在细胞浆内表达阳性率分别为 10 0 %、71%和 5 0 % ,p5 0、p65同时在胞核内表达 ,p5 0尤为明显 ,阳性率分别为 63%和 35 % ;5个乳腺癌细胞系细胞核MTT检测IC50 值大于 2 0 μg/ml,p5 0表达全部阳性 ,p65 2例阳性 ;p5 0胞核表达阳性者的IC50 值明显高于胞核表达阴性 ,而 p65无明显关系。 结论 :p5 0在乳腺癌细胞的发生中起一定作用 ,且与乳腺癌细胞的耐药关系密切 ,可望成为预后判断的新标志物

Abstract: | Objective:To study the expression of NFkB family proteins in breast cancer cell lines and its relationship with chemotherapy resistance of breast cell lines.Methods:Culture 14 breast cancer cell lines and extract protein from cytoplasm,nuclear separately;to detect the expression of NFkB family proteins and their inhibitor by western blotting and the relationship between the expression of NFkB family proteins and chemotherapy resistance MTT chemosensitivity testing.Results:In the expression of p65?p50 and IkB was in some of the cytoplasm the positive rate was 71%?100% and 50% and the positive rate reaches 35%?63% in nuclear.The IC 50 value of five breast caner cells was over 20μg/ml and their p50 expression was positive but only two cells appeared positive in p65.The IC 50 value of positive p50 expression was higher than that of negative p50 expression in nuclear and p65 had no similar relationship.Conclusion:p50 may play a role in the growth and developing of breast cancer cells and p50 is more remarkable; p50 had a close relationship with chemotherapy resistance of breast cancer cells and may be as a new mark to observe prognosis. |
