流式细胞仪检测网织红细胞百分比性能评价

目的:评价流式细胞仪测定外周血中网织红细胞(reticulocyte,Ret)百分比的性能。方法:应用流式细胞仪检测外周血Ret百分比,进行准确性、重复性及稳定性测试,并同时与手工计数结果比较,进行线性评价。结果:流式细胞仪检测外周血Ret百分比日间精密度与日内精密度CV均<0.36%,稳定性好;4℃低温24h内Ret百分比计数CV<0.43%,随着样本放置时间的延长,计数结果略有上升,不影响临床诊断;与同期手工法结果呈正相关关系(P<0.01)。结论:流式细胞仪测定Ret百分比准确性高,重复性、稳定性好,速度快,灵敏度高,纯度高,能满足临床需要,可替代手工计数。

Evaluation on performance of reticulocytes detection by flow cytometry

Objective：To evaluate the detection of reticulocytes by flow cytometry comprehensively.Methods：The percent of peripheral blood reticulocytes was detected by flow cytometry.The precision reproducibility and stability were observed.The results were compared with manual method and evaluated linearly.Results：Precision CV of the indexes was less than 0.36%,which had a good stability.And the percentage CV at 4℃ in 24 hours was less than 0.43%.With the prolongation of storage time of samples,the results increased slightly,but no effect on clinical diagnosis.It had a positive correlation with manual methods（P〈0.01）.Conclusions：Analyzing reticuloctyes by flow cytometry is a simple,fast,accurate,stable method and has a good reproducibility.It can meet clinical requirement and replace manual methods.
