miR-374a对肺腺癌细胞增殖与侵袭迁移的影响

目的探究分析miR-374a对非小细胞肺癌细胞增殖、侵袭、迁移等生物学行为能力的影响。方法采用RT-PCR检测miR-374a在正常肺上皮细胞CCD-8L及非小细胞肺癌细胞A549、H1975中的表达情况。采用miR-374a mimic和miR-374a inhibitor转染非小细胞肺癌细胞A549、H1975,采用RT-PCR检测各组细胞转染效率。分别采用CCK8细胞增殖实验、平板克隆实验、Transwell迁移实验和Transwell侵袭实验检测转染前后非小细胞肺癌细胞株增殖、迁移及侵袭能力变化情况。结果与正常肺上皮细胞CCD-8L比较,非小细胞肺癌细胞A549、H1975中miR-374a呈稳定高表达(P<0.05)。miR-374a mimic、miR-374a inhibitor可分别瞬时过表达、低表达非小细胞肺癌细胞A549、H1975中miR-374a表达情况(P<0.05)。过表达miR-374a可促进非小细胞肺癌细胞A549、H1975的增殖、迁移和侵袭(P<0.05);靶向抑制miR-374a表达可抑制非小细胞肺癌细胞A549、H1975的增殖、迁移和侵袭(P<0.05),组间差异显著,具有统计学意义。结论miR-374a在非小细胞肺癌细胞A549、H1975中呈稳定高表达,过表达miR-374a可明显促进非小细胞肺癌细胞A549、H1975的增殖、迁移和侵袭。

Effects of miR-374a on Proliferation,Invasion and Migration of Non-small cell Lung Cancer Cells

Objective To investigate and analyze the effects of miR-374a on the biological behavior of non-small cell lung cancer cells,including proliferation,invasion and migration.Methods RT-PCR was used to detect the expression of miR-374a in CCD-8L normal lung epithelial cells and A549 and H1975 non-small cell lung cancer cells.Non-small cell lung cancer cells A549 and H1975 were transfected with miR-374a mimic and miR-374a inhibitor,and the transfection efficiency of cells in each group was determined by RT-PCR.CCK8 cell proliferation assay,plate cloning assay,Transwell migration assay and Transwell invasion assay were used to detect the proliferation,migration and invasion of NSCLC cell lines before and after transfection.Results compared with CCD-8L in normal lung epithelial cells,miR-374a in A549 and H1975 of non-small cell lung cancer cells showed stable and high expression(P<0.05).miR-374a mimic and miR-374a inhibitor showed transient overexpression and low expression of miR-374a inhibitor in non-small cell lung cancer cells A549 and H1975,respectively(P<0.05).Over-expression of miR-374a promoted proliferation,migration and invasion of non-small cell lung cancer cells A549 and H1975(P<0.05).The targeted inhibition of miR-374a expression inhibited the proliferation,migration and invasion of non-small cell lung cancer cells A549 and H1975(P<0.05),and the difference between groups was significant and statistically significant.Conclusion miR-374a is highly expressed stably in NSCLC cells A549、H1975,and overexpression of miR-374a can significantly promote proliferation,migration and invasion of NSCLC cells A549、H1975.
