平肝通络颗粒对家兔脑膜血管的双相调节作用

背景药物治疗是偏头痛的主要治疗方法,而对平肝通络中药治疗偏头痛的作用机制的研究较少.目的证实平肝通络颗粒对家兔脑膜血管具有双相调节作用.设计完全随机安慰剂及阳性药物对照,单盲实验研究.地点和对象实验在湖南省中医药研究院动物室完成.采用家兔脑膜血管.市售家兔24只,雌雄不限,体质量2.5～3 kg,由湖南省中医药研究院动物室饲养2周以上.干预平肝通络颗粒十二指肠给药与静脉注射给药的颅通定对照,试验完成、血管管径恢复后,各再给药1次,30 min后由耳缘静脉按5μg/kg注射5-羟色胺.主要观察指标兔脑膜小血管管径值.结果平肝通络颗粒能够扩张正常家兔脑膜小血管管径,同时明显对抗注射5-羟色胺后兔脑膜血管管径的扩张,呈现双相调节作用.结论平肝通络颗粒治疗偏头痛有一定的药理学依据.

Dual-adjusting effect of the pinggantongluo granules on the small vessels of meninges in rabbits

BACKGROUND: Pharmacotherapy is the main treatment for migraine. However, few researches have been reported on the mechanism of treating migraine with traditional Chinese medicine ping gan ton gluo.OBJECTIVE: To confirm the dual-adjusting effect of the ping gan tong luo granules(PGTLG) on the meningeal small vessels in rabbits.DESIGN: A completely randomized, placebo-controlled, and single-blind experimental study was conducted.SETTING and PARTICIPANTS: The experiment was completed in the Laboratory Animal Room, Hunan Academy of Traditional Chinese Medicine. Rabbit meningeal vessels were used. Twenty-four rabbits, weighing 2.5 to 3 kg were purchased from markets with either gender, and kept for more than 2 weeks in the above research room.INTERVENTIONS: PGTLG was given to the duodenum, and intravenously administered tetrahydropalmatine was adopted as control. After the experiments were finished, PGTLG or control was given once more to restore vessel diameters; Thirty minutes later, 5 μg/kg of 5-HT was injected to the ear vein.MAIN OUTCOME MEASURES: Diameters of meningeal small vessels in rabbits.RESULTS: PGTLG dilated the meningeal small vessels in normal rabbits,and restrained the dilating of 5-TH-induced meningeal small vessels.CONCLUSION: PGTLG has a promising pharmacological effect in treating migraine.
