摘    要:目的探讨4种支原体宫内感染后孕鼠血清和羊水及子鼠血清IL-10、TNF-α、IL-1β、IL-6含量的变化,并予电镜观察子鼠心、脑和肺组织的变化,为支原体宫内感染导致不良妊娠的研究提供实验依据。方法取肺炎支原体(Mp)、发酵支原体(Mf)、梨支原体(Mpi)和穿通支原体(Mpe)分别感染正常妊娠组(31只)和不良妊娠组(23只)大鼠,在支原体宫内感染后,采用ELISA法检测孕鼠血清和羊水及子鼠血清IL-10、TNF—α、IL-1β、IL-6的含量,并对不良妊娠组子鼠心、脑和肺组织予电镜观察。结果Mp、Mf、Mpe及Mpi4种支原体宫内感染后,不良妊娠组子鼠血清和孕鼠羊水TNF—α、IL-1β、IL-6水平均较正常妊娠组显著增高(均P〈001),而子鼠血清和孕鼠羊水IL-10水平则较正常妊娠组显著降低(均P〈001);同时发现,不良妊娠组Mf和Mp感染的子鼠肺组织内均可见组织水肿和炎症反应,而Mpi感染的子鼠脑组织内可见明显组织损伤,Mpe感染子鼠的心脏组织中可见三层膜结构的支原体颗粒、线粒体固缩、肌浆网扩张、核膜间隙扩大和局灶性肌丝溶解现象。结论Mp、Mf、Mpe及Mpi4种支原体宫内感染可导致不良妊娠。

Ultrastructural changes of the heart,brain and lung tissues and cytokines levels in neonatal rats with intrauterine infection of four mycoplasmas

Abstract:Objective To investigate cytokines levels in the serum and amniotic fluid in pregnant rats with intrauterine infection of four mycoplasmas and their newborns, and to observe the ultrastructural changes of heart, brain and lung tissue in the neonatal rats. Methods The pregnant rats were infected with Mycoplasma pneumoniae (MP), Mycoplasma fermentans (Mf), Mycoplasma penetrans (MPe) and Mycoplasma pirum (MPi), respectively. After intrauterine Mycoplasma infection, the serum and amniotic fluid levels of IL-1β, IL-6, TNF-α and IL-10 in neonatal and pregnant rats were measured with ELISA. The heart, brain and lung tissues were observed with electron microscope in the neonatal rats of adverse pregnancy group. Results After intrauterine infection with four mycoplasmas, TNF-α, IL-1β and IL-6 levels in serum of neonatal rats and amniotic fluid of pregnant rats were increased significantly (P〈0.01); but those of IL-10 in adverse pregnancy group were significantly lower than in normal pregnancy group (P〈0.01). Ultrastructural observation indicated that there was inflammatory damage in the heart, brain and lung of the neonatal rats. Conclusion The intrauterine infection with Mp, Mf, Mpe and Mpi can result in varied adverse pregnancy outcomes in rats.
