摘    要:目的:建立神经干细胞分离、培养的技术方法,探讨表皮生长因子(EGF)对大鼠胚胎神经干细胞增殖、分化的影响。方法:从大鼠胚胎大脑组织中分离、培养神经干细胞,并用EGF和血清诱导其分化,采用免疫细胞化学方法鉴定神经干细胞增殖和分化的神经样细胞。结果:大鼠胚胎神经干细胞在无细胞因子和血清的培养基中无新生细胞形成,但能在EGF和血清的诱导下产生nestin和BrdU阳性细胞,细胞贴壁后分化为神经元和星形胶质样细胞。结论:EGF和血清能促进神经干细胞增殖,并诱导其向神经元样和星形胶质样细胞分化。

Abstract:Aim:To establish the methods of isolation and culture of neural stem cells and investigate the effects of epidermal growth factor (EGF) on proliferation and differentiation of rat embryonic neural stem cells in vitro.Methods: Neural stem cells separated from embryonic rat brains were cultured in the medium containing EGF and serum. Immunocytochemistry were applied to identify neural stem cells and their derivative neuron like and glia like cells.Results: The rat embryonic neural stem cells showed no ability of self renew in medium without EGF and serum. However they could become nestin and BrdU positive cells and differentiated into neuron like cells and astrocyte like cells in medium containing EGF and serum after adherence.Conclusion:EGF and serum induce rat embryonic neural stem cells to proliferate and differentiate into neuron like and astrocyte like cells in vitro.
