枳黄方对酒精性肝损伤大鼠肝胃ADH1 mRNA的影响

目的:探讨枳黄方对大鼠酒精性肝损伤的防护作用及其机理。方法:将30只SD大鼠随机分为3组:空白对照组、酒精攻击组、枳黄方防护组,每组10只。10天后,处死大鼠,取大鼠肝脏进行HE染色观察肝病理变化,RT-PCR法测定肝、胃组织ADH1mRNA的变化。结果:枳黄方能缩短大鼠醉酒时间(P<0.05),枳黄方防护组肝、胃病理表现较模型组明显改善,其肝、胃ADH1mRNA表达量高于模型组(P<0.01)。结论:枳黄方对大鼠酒精性肝损伤有较明显的防护作用,这可能与其能提高肝、胃ADH1mRNA的表达有关。

Effect of Zhihuang decoction on liver and stomach ADH1 mRNA expression in rats with alcohol-induced liver disease

Objective:To study the protective effect of Zhihuang decoction on alcohol-induced liver disease in rat and its mechanism.Methods:Thirty SD rats were randomly divided into three groups:control group,model group and the Zhihuang decoction treatment group.56% ethanol in the dose of 8g/kg body weight was intragastricly administrated to 20 rats for ten days to make the model of alcohol-induced liver disease,and ten of these 20 rats were treated with Zhihuang decoction simulataneously.All rats were sacrificed ten days later.The pathological changes in the liver were observed under light microscope with hematoxylin and eosin(HE) staining.RT-PCR method was employed to detect the expression of ADH1mRNA in liver and stomach.Results:Zhihuang decoction could shorten the drunkenness time of rats(P<0.05);It could alleviate the pathologic change of liver and stomach;It also enhanced to expression of ADH1mRNA in liver and stomach(P<0.01),compared with that of model group.Conclusion:Zhihuang decoction has certain protective effect on hepatic injure induced by alcohol in rats by enhancing the expression of ADH1mRNA in liver and stomach.
