摘    要:目的 探讨藻酸盐万古霉素缓释微球的制备与优化.方法 将不同分子结构和理化特性的藻酸盐不同配比,应用滴注法制备不同浓度万古霉素藻酸盐微球,经过冻干、低压冻干直至重量不再变化,取样微球进行裂解,取上清液在高效液相色谱仪上测定万古霉素浓度,并按公式计算包封率,对各组所得包封率数值和微球颗粒大小结果进行统计观测.结果 不同种类...

Abstract:Objective To explore the preparation and optimization of vancomycin alginate beads.Methods Different concentration of alginate vancomycin palliative beads were prepared using titration in various proportion according to their different molecular structure and physio-chemical properties.After continuous low-pressure lyophilization till the weight did not changed,sample beads were collected and disrupted with supernatant subjected to HPLC analysis,and encapsulation rate was calculated.Both encapsulated rate and diameter of beads were subjected to statistical analysis.Results The initial comparison of encapsulation rates of multiple concentrations and kinds of alginate sodium demonstrated that encapsulation rates increased with higher concentration of alginate sodium with high G and elasticity.Beads diameter showed minimal increase with the elevation of elasticity and concentration.The encapsulation rate of alginate vancomycin beads with the G∶H ratio of 2∶1 was significantly better that other groups.Conclusion The optimal formulation of alginate vancomycin palliative beads was obtained for further optimization and future in vivo osseous tissue engineering experiments in animals.
