黄荆子乙酸乙酯提取物对人肝癌裸鼠移植瘤生长的影响

摘    要:目的:观察黄荆子乙酸乙酯提取物(EVn-50)对人肝癌Hep G2裸鼠移植瘤生长的抑制作用。方法:采用人肝癌Hep G2裸鼠模型评估EVn-50在体治疗肝癌的有效性;免疫组化染色检测增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、血管内皮生长因子(vascular endothelial growth factor,VEGF)、CD31蛋白表达。结果:EVn-50显著抑制人肝癌Hep G2裸鼠移植瘤的生长,呈剂量依赖性;5,10,20 mg/kg的EVn-50对移植瘤的瘤重抑制率分别为55%,62%,69%。免疫组化染色结果显示,EVn-50(5,10,20 mg/kg)与生理盐水组比较,PCNA、VEGF、CD31的表达下调,亦呈剂量依赖性。结论:EVn-50具有抑制人肝癌HepG2裸鼠移植瘤生长作用,其作用机制可能与其下调PCNA、VEGF、CD31的表达有关。

Abstract:Objective To investigate the effects of the acetoacetate extract of Vitex negundo seed(EVn-50) on the growth of human hepatocellular carcinoma xenograft in nude mice.Methods Human hepatocellular carcinoma xenograft in nude mouse model was established to evaluate the therapeutical efficacy of EVn-50 in vivo.The expression of proliferating cell nuclear antigen(PCNA),vascular endothelial growth factor(VEGF) and CD31 in the 20 xenograft specimens were detected by immunohistochemistry.Results Therapeutic trial of human hepatocellular carcinoma xenograft in nude mice showed that EVn-50 obviously inhibited the growth of human hepatocellular carcinoma xenograft,in a dose-dependent manner.The tumor weight inhibitory rate of EVn-50(5,10,20mg/kg) was 55%,62%,69%,respectively.Data of immunohistochemistry indicated that the expression of PCNA,VEGF and CD31 all decreased after treatment with EVn-50(5,10,20mg/kg),in a dose-dependent manner.Conclusion EVn-50 possess therapeutic action of human hepatocellular carcinoma xenograft in nude mice,and its mechanism is involved in the down-regulation the protein expression of PCNA,VEGF and CD31.
