扶正抑瘤汤对肿瘤细胞周期及端粒酶影响的实验研究

摘    要:目的:探讨扶正抑瘤汤抗肿瘤的作用机理。方法:用S180瘤株接种昆明种小鼠,造成S180实体瘤模型,扶正抑瘤汤等中药灌胃治疗,用流式细胞仪分析实体瘤的细胞周期;用端粒酶试剂盒检测端粒酶活性。结果:各治疗组肿瘤细胞G0/1期比率升高,S期比率下降,端粒酶活性被抑制,与生理盐水(荷瘤)组相比,差异有显著性(P<0.001)。结论:扶正抑瘤汤能阻滞肿瘤细胞增殖的进程,阻抑肿瘤细胞DNA的合成和复制,而端粒酶活性被抑制是影响肿瘤细胞增殖周期进程的机理之一。

Experimental Study on Effect of Fuzheng Yiliu Decoction on Tumor Cell Cycle and Telomerease

Abstract:To study the mechanism of anti tumor effect of Fuzheng Yiliu Decoction (FZYLD). Methods: S 180 neoplasm strain was inoculated in Kunming mice to establish model of S 180 solid tumor. The model animals were treated with FZYLD by gastrogavage, the cell cycle of tumor were checked up by flow cytometer and the telomerease kit was used to test telomerease activity. Results: The stage G 0/1 ratio of tumor cells in model animals treated with FZYLD increased, while cells of S stage decreased, with telomerease activity inhibited. These changes were different significantly from those in the model animals treated with normal saline (P<0 001). Conclusion: FZYLD could block the tumor cell proliferation procedure and inhibit the DNA synthesis and duplication in tumor cell. And the suppression of telomerease activity might be one of the mechanisms affecting the tumor cell proliferation cycle.
