兔眼球破裂伤视网膜细胞内RNA表达与凋亡

目的 观察兔眼球破裂伤后视网膜细胞内RNA的表达和凋亡情况.方法 健康新西兰白兔制作眼球破裂伤模型,用甲基绿-派罗宁-马休黄（MG-P-MY）染色法行细胞RNA检测,TUNEL法细胞原位凋亡检测.结果 伤后3 h组在内核层及神经节细胞层有少量RNA阳性表达;6 h内核层阳性表达增多;12 h阳性表达明显;24 h可见大量RNA阳性细胞.伤后3 h组视网膜细胞中偶见凋亡细胞;6 h组凋亡细胞显著增加;12 h～24 h组见大量凋亡细胞.结论 眼球破裂伤后视细胞内早期即有RNA表达,TUNEL法检测存在凋亡现象.

The RNA and TUNEL positive cells of retina after the eyeball rupture

Objective To investigate the RNA and TUNEL positive cells of retina after the rabbit eyeball rupture. Methods White New Zealand rabbits were selected to do eyeball rupture model,MG-P-MY stain was used to examine RNA and TUNEL was used to locate apoptosis respectively. Results In MG-P-MY stain, the RNA had no obvious expression in the control group and while in experimental group,3 hours later, a lower positive expression of RNA were observed in the ganglion and inner layer cells, it increased obviously in 6 hour groups and there were more RNA in 12 hour groups, numerous of RNA expression cells could be seen in 24 hour groups. In TUNEL stain,the apoptosis had lower expression in ganglion cells and the inner and outer nuclear layer 6 hours later,an obvious increasing of dye positive cells were observed between 12 and 24 hours. Conclusions There is RNA expression early in retina cells after injury and the apoptosis cells can be seen by TUNEL stain.
