摘    要:目的 探究过表达亮氨酸拉链假定肿瘤抑制基因1(LZTS1)对胰腺癌PANC-1细胞的增殖、凋亡、侵袭、迁移能力的影响.方法 将PANC-1细胞按照随机数字表法分为四组,对照组(常规培养)、NC组(转染阴性对照)、pcDNA-LZTS1-1组(转染pcDNA 3.0 LZTS1-1质粒)、pcDNA-LZTS1-2组(转染pcDNA 3.0 LZTS1-2质粒).蛋白质印迹法(Western blotting)检测转染效果;噻唑蓝(MTT)观察PANC-1细胞增殖,流式细胞术检测凋亡率,Transwell法检测细胞的侵袭、迁移.结果 与对照组相比,pcDNA-LZTS1-1组和pcDNA-LZTS1-2组PANC-1细胞中LZTS1蛋白水平[(2.835±0.301),(4.125±0.385)比(1.000±0.085)]显著增加,其中pcDNA-LZTS1-2组增加显著;过表达LZTS1抑制PANC-1细胞增殖、侵袭[(56.369±6.432)个比(117.258±12.152)个]、迁移[(120.145±14.210)个比(233.258±25.148)个],诱导其凋亡[(23.252±2.152)%比(6.589±0.645)%].结论 过表达LZTS1显著抑制PANC-1细胞增殖、侵袭、迁移,促进其凋亡.

Abstract:Objective To investigate the effect of overexpression leucinezipper putative tumor suppressor 1 (LZTS1) on the proliferation, apoptosis, invasion and migration ability of pancreatic cancer PANC-1 cells.Methods PANC-1 cells were randomly divided into4 groups according to random number table method: control group (conventional culture), NC group (transfected with negative control),pcDNA-LZTS1-1 group (transfected with pcDNA 3.0 LZTS1-1) and pcDNA-LZTS1-2 group (transfected with pcDNA 3.0 LZTS1-2). The transfection effect was detected by Western blotting. The proliferation of PANC-1 cells was observed by MTT and apoptotic ratewas detected by flow cytometry. The cell invasion and migration were detected by Transwell assay.Results Compared with the control group, the level of LZTS1 protein [(2.835±0.301), (4.125±0.385) vs. (1.000±0.085)] of PANC-1 cells in the pcDNA-LZTS1-1 group and the pcDNA-LZTS1-2 group was significantly increased, especially in the pcDNA-LZTS1-2 group. Overexpression of LZTS1 significantly inhibited the proliferation, invasion [(56.369±6.432) vs. (117.258±12.152)] and migration [(120.145±14.210) vs. (233.258±25.148)] of PANC-1 cells and induces apoptosis [(23.252±2.152)% vs. (6.589±0.645)%].Conclusion Overexpression of LZTS1 significantly inhibits the proliferation, invasion and migration of PANC-1 cells and promotes apoptosis.
