C57BL/6J小鼠脂肪细胞抵抗胰岛素抑制脂解的分析

摘    要:为了从细胞水平探讨C57BL/6J和A/J小鼠对胰岛素(Ins)的敏感性,该实验分离了这两种小鼠的脂肪细胞,观察Ins 抑制脂肪细胞在异丙肾上腺素介导下的脂解作用,以甘油的释放量作为脂解的指标。结果表明:C57BL/6J的脂肪细胞在Ins 400和800 pm ol时甘油的释放量多于A/J(P< 0.01),即Ins抗脂解能力显著低于A/J小鼠,提示C57BL/6J小鼠脂肪细胞抵抗Ins抑制脂解。这与C57BL/6J为什么易于发生2 型糖尿病可能存在病理生理方面的联系。

Abstract:It has been demonstrated that C57BL/6J mice is prone to develop non insulin dependent diabetes mellitus (NIDDM),but A/J mice isn't.NIDDM is related to insulin resistance.However,it is unknown whether C57BL/6J mice exist insulin resistance before developing NIDDM.In order to investigate insulin sensitivity from cellular level,we assessed the effect of insulin inhibiting the isoproterenol induced lipolysis in adipocytes isolated from C57BL/6J and A/J mice.Glycerol released from adipocyte medium was used as the index of lipolysis,Glycerol concentration was significantly higher in adipocytes from C57BL/6J than that from A/J at 400 and 800 pmol insulin in medium( P <0 01).That meant the ability of insulin inhibiting lipolysis was lower in adipocyte of C57BL/6J than in that of A/J.Resistance to the antilipolytic action of insulin in adipocyte from C57BL/6J may be associated with the susceptibility to develop NIDDM of C57BL/6J mice in pathophysiology.
