非小细胞肺癌nm23-H1等位基因缺失与淋巴结转移关系的研究

摘    要:目的探索非小细胞肺癌中nm23-H1等位基因缺失(LOH)与淋巴结转移的关系.方法 Southern印迹杂交检测39例非小细胞肺癌组织及相应癌旁正常肺组织中nm23-H1等位基因缺失的情况,并探索其与非小细肺癌淋巴结转移的关系.结果 nm23-H1两条等位基因分别为7.6、2.3 kb,伴有淋巴结转移的肺癌nm23-H1等位基因缺失发生率为48%(12/25),高于不伴淋巴结转移者8%(1/11),差异有显著统计学意义(P<0.05).结论 nm23-H1等位基因缺失可能诱发非小细胞肺癌淋巴结转移,与其淋巴结转移关系密切,检测nm23-H1等位基因缺失可作为判断非小细胞肺癌淋巴结转移有用的基因标志物之一.

Abstract:Objective To explore the relationship between the loss of the heterozygosity of nm23-H_1 and the lymph node metastasis in no-small-cell lung cancer (NSCLC). Methods The loss of the heterozygosity of nm23-H_1 gene of cancer tissue and peri-cancerous tissue was detected in 39 patients with NSCLC by Southern blotting. Results The heterozygosity gene of nm23-H_1 is 7.6 kb and 2.3 kb. The LOH rate of the patients with lymph node metastasis (48%, 12/25) was higher than those without lymph node involvement (8%, 1/11) (P<0.05). Conclusion The loss of heterozygosity of nm23-H_1 is related to the metastasis of lymph node. It can be used as an important gene marker to predict the metastasis of lung carcinoma via lymph path.
