摘    要:目的探讨表没食子儿茶素没食子酸酯(EGCG)对人肝癌细胞株SMMC-7721生长的影响及其可能作用机制。方法采用不同浓度的EGCG处理SMMC-7721细胞,以四唑氮蓝还原法(MTT)观察处理后细胞的生长情况,流式细胞术(FCM)检测细胞凋亡情况,逆转录聚合酶链式反应(RT-PCR)检测抑癌基因PUMA mRNA表达水平的变化,Western-Blot检测PUMA蛋白水平的表达。结果 EGCG处理后SMMC-7721细胞生长明显抑制,并且呈现时间、剂量依赖性,凋亡细胞比例明显上升。抑癌基因PUMA的mRNA水平和蛋白表达水平随着EGCG浓度的增加而升高。结论 EGCG可以明显抑制人肝癌SMMC-7721细胞的生长,诱导其凋亡;其可能机制为上调抑癌基因PUMA的表达水平,从而促进细胞凋亡。

Abstract:Objective It is to approach the effect of epigallocatechin-3-gallate(EGCG) on proliferation of human hepatoma carcinoma cell line SMMC-7721 and the underlying mechanism.Methods Human hepatoma carcinoma cell SMMC-7721 was treated with different concentrations of EGCG;the proliferation of cells was assayed by MTT method;flow cytometry(FCM) was used to detect cell apoptosis;the expression of PUMA mRNA was analyzed by RT-PCR,while PUMA protein expression was determined by Western-blot.Results Treatment of EGCG led to a significant inhibition on proliferation of SMMC-7721 cells in a dose-and time-dependent manner.The percentage of apoptotic cells increased markedly after EGCG treatment.In this process,both mRNA and protein expression of PUMA gene were elevated.Conclusion EGCG can effectively inhibit the proliferation of SMMC-7721 cells.The underlying mechanism may due to the up-regulation of PUMA,which lead to an increased apoptosis.
