人参皂苷Rg1、Rb1对淀粉样前体蛋白分泌酶代谢途径的影响

摘    要:目的观察人参皂苷Rg1、Rb1对淀粉样前体蛋白代谢途径中α分泌酶活性和基因表达的影响。方法用转染突变型PSl(ML46L)和野生型PSl的CHO细胞,通过荧光酶标技术检测α分泌酶活性,Westernblot免疫印迹、RT-PCR技术检测α分泌酶基因ADAM9、ADAM10的蛋白和RNA表达。结果转染突变型PSlML46L组a分泌酶活性较转染野生型PS1(WT组)低(P〈0.05),加用人参皂苷Rg1、Rb1后转染突变型PSlML46L组α分泌酶活性较未加用组不同程度增高(P〈0.05),同时ADAM9、ADAM10的蛋白及RNA表达较未加用组有不同程度增多(P〈0.05)。结论人参皂苷Rg1、Rb1均可不同程度增加α分泌酶中ADAM9、ADAM10基因表达,提高α分泌酶活性,且人参皂苷Rg1调节作用优于人参皂苷Rb1。
