黑木耳粉对矽肺大鼠体内抗氧化水平的影响

摘    要:目的:探讨黑木耳粉对染矽尘大鼠体内抗氧化物酶活性和脂质过氧化水平的影响。方法:将40只Wister大鼠随机分为阴性对照组、二氧化硅(SiO2)对照组、黑木耳粉高、中和低剂量组,每组8只。采用非暴露式气管插管方法给实验组大鼠染SiO2粉尘,阴性对照组大鼠给予生理盐水,24 h后黑木耳粉干预各剂量组给予黑木耳粉饲料喂养。于染尘后28 d处死大鼠,测定肺组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量,测定肺泡灌洗液(BALF)中乳酸脱氢酶(LDH)活性,采用HE染色观察肺组织的病理变化。结果:28 d时,与SiO2对照组相比,黑木耳粉各剂量组SOD活性升高,差异有统计学意义;黑木耳粉高剂量组GSH-Px活性较SiO2对照组升高,差异也具有统计学意义;黑木耳粉高、中剂量组MDA的含量及LDH活性与SiO2对照组相比降低,差异均具有统计学意义;病理形态学观察显示,黑木耳粉各剂量组大鼠肺脏损伤程度较SiO2对照组有所减轻。结论:黑木耳粉具有提高矽肺大鼠体内抗氧化水平的作用,在一定程度上能够延缓或阻抑矽肺病变的发展。

Abstract:Objective: To explore the effects of Auricularia auricular powders(AAP) on antioxidase activities and the level of lipid peroxidation in rat lungs exposed to silica.Methods: Forty rats were randomly divided into the control group,silica group and 3 AAP-treated groups(large dose group,moderate dose group and small dose group),8 for each.Animal models were established by direct tracheal instillation of silica into rat lungs non-surgically.24 hours after the establishment of the animal model,rats in AAP-treated groups were given AAP by feeding.On the 28th day,the content of MDA,the activity of SOD,GSH-Px and LDH were assayed.Lung fibrosis in morphology was observed by H.E staining.Results: On the 28 th day,the activity of GSH-Px and SOD of the rats in AAP-treated groups was significantly higher than that in SiO2 group.The content of MDA of the rats in AAP-treated groups was significantly lower than that in SiO2 group.The activity of LDH of the rats in AAP-treated groups was lower than that in SiO2 group.The fibortic degree of rats lung was lighter in AAP-treated groups than that in SiO2 group.Conclusion: AAP possesses the ability of enhancing the antioxidation level of rat lungs exposed to silica and can partly restrain or inhibit the development of silicosis.
