| Abstract: | OBJECTIVE To establish the method for the determination of phenytoin in serum by HPLC.METHODS HPLC was used C18 column(4.6mm×150mm,5μm) was adopted,and the temperature was set at 30℃,The UV detector was operated at 220nm,mobile phase of methanol-acetonitrile-water(10∶30∶60),and the flow rate was 1mL·min-1.RESULTS The calibration curve was linear in the concentrations range from 5 to 40μg·mL-1(R=0.9993172).Within the range,the method recovery rates of low、moderate and high concentrations were 101.76%、100.48%、100.80%,RSD was 2.36%(intra-day) and 4.95%(inter-day).CONCLUSION The method is convenient、rapid,suitable for TDM. |
