高效液相色谱法测定对乙酰氨基酚颗粒的溶出度

摘    要:目的建立对乙酰氨基酚颗粒溶出度的高效液相测定方法。方法色谱柱为Waters C18柱(4.6mm×150mm,5μm),流动相为磷酸盐缓冲液(pH4.5)-甲醇(80:20),流速为1.0mL·min^-1,检测波长为254nm,柱温为30℃。结果对乙酰氨基酚在0.05-0.40mg·mL^-1浓度范围内呈良好线性关系(r=1.0),平均回收率为99.9%(n=9),RSD为0.27%。结论该方法准确、可靠,与标准法比较,结果较为满意,可用于对乙酰氨基酚颗粒的溶出度测定。

Abstract:OBJECTIVE To establish a HPLC method for dissolution of Paracetamol Granules.METHODS Waters C18 column(4.6mm×150mm,5μm)was used with a mobile phase composed of phosphate bufering solution(pH4.5)-methanol(80∶20)at the flow rate of 1.0mL·min-1,the detection wavelength was 254 nm and the temperature of the column Was 30℃.RESULTS The calibration curve of paracetamol was linear in the range of 0.05~0.40mg·mL-1,r=1.0.the average recovery rate Was 99.9% with a RSD of 0.27%(n=9).CONCLUSION This method is accurate,realiable and can be used for the of dissolution Paracetamol Granules.
