血府逐瘀胶囊对单侧输尿管梗阻大鼠Smad信号通路分子的影响

摘    要:目的探讨血府逐瘀胶囊对单侧输尿管梗阻(unilateral ureteral obstruction,UUO)所致大鼠肾间质纤维化的作用及机理。方法雄性SD大鼠34只随机分为4组:正常组(6只)、假手术组(6只)、模型组(11只)、血府逐瘀胶囊治疗组(11只),采用UUO法建立大鼠肾间质纤维化模型,造模次日予以血府逐瘀胶囊灌胃治疗,3周后检测血清肌酐和尿素氮含量,观察肾功能变化;取肾组织进行HE和六胺银染色(periodic acid-silver metheramine,PASM),观察肾组织病理变化;采用Western blotting分别检测肾组织中磷酸化Smad2(phospho-Smad2,p-Smad2),Smad2,Smad4,Smad7蛋白表达水平,观察Smad信号通路分子表达水平的变化。结果与正常组和假手术组比较,模型组大鼠血肌酐和尿素氮含量显著上升;肾小管基底膜明显增厚,部分肾小管上皮细胞变性,肾小管扩张与萎缩并存,肾间质明显增宽,纤维组织增生,大量炎细胞浸润;p-Smad2和Smad2蛋白表达显著上调,Smad7蛋白表达显著下调(P<0.01),而Smad4蛋白表达水平无明显差异。各项指...
