摘    要:目的 研究刺五加 (AS)对受谷氨酸毒性作用的神经元的保护作用。方法 用大鼠离体海马脑片和组织学检查 ,观察使用AS注射液组 (AS组 )和未使用AS组 (对照组 )对谷氨酸 (Glu)所致的缺血海马脑片顺向群峰电位 (OPS)的影响 ,及两组脑片的超微结构变化。结果 AS组脑片在加入Glu后 5分钟左右OPS减小并消失 ,去除Glu 1小时后OPS恢复率为 83.3% ,平均恢复程度为 86 .4 % ;对照组脑片加入Glu后OPS在 3分钟左右迅速减小并消失 ,去除Glu 1小时后OPS恢复率为 16 .6 % ,恢复程度为 4 1.5 %。两组比较有显著性差异 (P <0 .0 5 )。电镜下观察发现 ,加有Glu脑片的CA1区锥体细胞核染色加深 ,核膜不完整 ,核内染色质凝聚成块 ,胞浆中内质网高度扩张。使用AS后的神经细胞核膜完整 ,核内染色质轻度凝聚 ,内质网轻度扩张。结论 AS对Glu毒性作用所致的脑损伤具保护作用 ,该作用机制可能是AS能减轻因缺血缺氧所致的神经元损伤

Abstract:Objective To study the protective effect of Acathopanax Senticosus(AS) to the neuron by toxicity of Glu.Methods With hippocampal slice in rats and histography technique,we observed the effect in OPS of ischemic hippocampal slice by Glu between AS group and no AS group,and the changes of ultrastructure in the two groups were also observed.Results In AS group OPS was decreased and disappeared after slice with Glu for 5 min,the recovery rate of OPS was 16.6%,the degree was 41.5%.There was significant difference in the two groups( P < 0.05 ).With electromicroscope we found the pyramidal neurons in CA 1 region were damaged by Glu exposure, the nucleus membranes weren't complete.the chromatin formed mass.the endoplasmic in cytoplasm expanded,the nucleus membranes of the nerve cells were complete after using AS,the chromatin in the nucleus of cells condensed,the endoplasmic reticulum expanded lightly.Conclusions AS could protect hippocampal neurons from the excitatory toxicity of Glu. Its mechanism is probably AS can lighten neuronal injury by anoxic ischemia.
