摘    要:目的:优化淡豆豉中大豆异黄酮的高效液相色谱含量测定方法。方法:采用色谱柱为Agilent Hyper-sil C18(4.0 mm×250 mm 5,μm),流动相为甲醇-0.09%醋酸(48:52),流速1.0 mL.min-1,测定波长为254nm,检测温度为25℃的色谱条件测定。结果:线性范围:大豆苷40.6~203μg(r=0.9998),染料木苷11.15~55.75μg(r=0.9926),大豆苷元12.05~60.25μg(r=0.9933),染料木素5.5~27.5μg(r=0.9946)。测定精密度:大豆苷RSD=1.59%,染料木苷RSD=1.7%,大豆苷元RSD=0.8%,染料木素RSD=1.6%。大豆异黄酮总量24h稳定性RSD=1.49%,重复性RSD=1.06%,加样回收率:大豆苷为(99.24±3.77)%,染料木苷为(99.66±5.82)%,大豆苷元为(99.41±4.33)%,染料木素为(99.06±7.09)%。结论:本法简便、可靠、灵敏,可作为大豆异黄酮的质量控制方法。

Abstract:Objective: To improve the HPLC method for the content determination of soy isoflavon from Semen Sojae praeparatum.Method: Isoflavone was analyzed on a Agilent Hypersil C18 column(4.0mm×250mm,5μm).A mixture of MeOH-0.09 % HAc(48∶ 52) was used as the mobile phase with a flow rate of 1.0mL·min-1.The detecting wavelength was 254 nm at 25℃.Result: The quantitative linear ranges of four isoflavones were daidzin 40.6~203μg(r=0.9998),11.15~55.75μg(r=0.9926),daidzein 12.05~60.25μg(r=0.9933),genistein 5.5~27.5μg(r=0.9946),the precisions were RSD=1.59%,RSD=1.7%,RSD=0.8%,RSD=1.6%,the stability of isoflavonoids in 24h was RSD=1.49%,reproducibility RSD=1.06%.The recoveries of daidzin was(99.24±3.77)%,genistin was 99.66±5.82%,daidzein was(99.41±4.33)%,genistein was(99.06±7.09)%.Conclusion:The method is convenient,reliable and sensitive so it can be used for the quality control of soy isoflavone.
