模拟中医病因所致脾气虚、脾阳虚大鼠模型的证候比较研究

目的:以多因素复合法分别建立脾气虚证、脾阳虚证大鼠模型,对两组模型大鼠进行比较研究以探讨脾气虚、脾阳虚的证候实质与差异。方法:动物随机分为正常组、脾气虚组和脾阳虚组,以相应造模方法进行模型复制,并以四君子汤和理中汤进行药物反证。结果:脾气虚和脾阳虚大鼠宏观证候计分高于正常组(P0.01),且以脾阳虚组为著(P0.01);两组血清GAS均低于正常组(P0.01);脾气虚组的血浆MTL低于正常组(P0.01),而脾阳虚组高于正常组(P0.01);经健脾中药干预后,各项脾虚症状和体征逐渐缓解或消失;血清GAS、血浆MTL紊乱回复。结论:多因素复合法复制的脾气虚、脾阳虚证大鼠模型符合证候的生物学特征,稳定性良好,且经得起药物反证;脾气虚、脾阳虚大鼠的脾虚宏观证候呈递进关系;血浆MTL在脾气虚和脾阳虚中变化趋势不一致,可作为鉴别两证的微观指标。

Comparative Study on Model Rats with Spleen-qi Deficiency Syndrome and Spleen-yang Deficiency Syndrome

Objective: By making a comparative study on rats with spleen-qi deficiency syndrome and spleen-yang deficiency syndrome,to seek the objective property of these two syndromes and their differences.Methods: Animals were randomly divided into three groups in terms of control group,group of spleen-qi deficiency syndrome and spleen-yang deficiency syndrome.Model rats were induced by combination of multi-methods and testified with corresponding medicinals(with Sijunzi Decoction for spleen-qi deficiency syndrome and Lizhong Decoction for spleen-yang deficiency syndrome).Results: Scores of symptoms and signs in two model groups were significantly higher than those in control group(P<0 01),and the difference between two model groups was significant as well,with group of spleen-yang deficiency higher(P<0 01).Serum GAS was significantly decreased in model groups(P<0 01).Plasma MTL was significantly decreased in group of spleen-qi deficiency(P<0 01),while increased in spleen-yang deficiency(P<0 01).After being treated with medicinals,the indexes mentioned above were recovered somehow.Conclusion: Multi-methods applied in this research to induce spleen-qi deficiency syndrome and spleen-yang deficiency syndrome are proved to be feasible and can be popularized.There is a calculable progressing in symptoms and signs between spleen-yang deficiency and spleen-qi deficiency.The variation tendency of plasma MTL in spleen-qi deficiency and spleen-yang deficiency is inconsistent,which may be regarded as an objective index to make syndrome differentiation.
