摘    要:目的:建立连翘药材中连翘苷的含量测定方法,比较不同产地连翘中连翘苷的含量。方法:采用反相高效液相色谱法测定不同产地连翘药材中连翘苷的含量,流动相为乙腈-水(25∶75),流速为1.0mL.min-1,检测波长为277nm,进样量为10μL,柱温为30℃。结果:在选定色谱条件下,连翘苷分离度良好,线性范围为0.4348~4.348μgr,=0.9999,平均回收率98.9%,RSD=0.68%(n=6);不同产地连翘药材中连翘苷含量差异较大,其中陕西洛南,湖北邵县以及四川成都的连翘药材中连翘苷含量相对高于其它产地。结论:该方法测定连翘中连翘苷的含量,简单、灵敏、准确、重现性好,连翘苷在各产地连翘间差异较大。

Abstract:Objective: To develop a method for determining the content of phillyrin in Fructus Forsythiae and to compare the content in Fructus Forsythiae from different habitats.Methods:HPLC was adopted to determine the content of phillyrin,the mobile phase was acetonitrile-water(25∶ 75),the velocity was 1.0 mL·min-1,the UV detection wavelength was 277nm,the sampling volume was 10mL,the temperature of column was 30℃.Results: Under the selected Chromatography condition the phillyrin was observed in a good separation degree,a good linear with range of 0.4348~4.348μg,r=0.9999,the average recovery was 98.9% and RSD was 0.68%(n=6).The content of phillyrin in Fructus Forsythiae from different habitats is different,of which the content of phillyrin in Fructus Forsythiae from Luonan(Shaanxi),Shaoxian(Hubei) and Chengdu(Sichuan) are higher than that of others.Conclusion:The method is simple,sensitive,accurate and with good reproducibility.It can be used to determine the content of phillyrin in Fructus Forsythiae.The content is different with the habitat of Fructus Forsythiae.
