升清降糖合剂对糖尿病大鼠胰岛细胞形态及功能的影响

摘    要:目的:探讨升清降糖合剂(简称SQ)对糖尿病大鼠空腹血糖水平和胰岛细胞形态及功能的影响。方法:腹腔注射链脲佐菌素(STZ)诱导建立糖尿病大鼠模型。根据不同的干预方式将糖尿病大鼠分为模型对照组、SQ低剂量组、SQ中剂量组、SQ高剂量组。每两周测空腹血糖和体重,于干预后第八周末,各组大鼠麻醉后采集血样,ELISA法测定血清C肽水平,取胰腺组织HE染色观察病理学改变。结果:空腹血糖水平,模型对照组(26.92±3.68)SQ低剂量组(23.87±2.86)SQ高剂量组(20.66±2.81)SQ中剂量组(20.66±2.81)正常对照组(4.55±0.57),SQ中、高剂量与模型对照组比较差异有统计学意义(P0.05)。血清C肽水平,正常对照组(2.27±0.47)SQ中剂量组(1.76±0.91)SQ高剂量组(1.74±0.79)SQ低剂量组(1.11±0.36)模型对照组(0.70±0.62),SQ中、高剂量组与模型对照组比较差异有统计学意义(P0.05)。胰岛病理切片形态学观察发现,模型对照组大鼠胰岛细胞数量减少,排列紊乱,胞质呈空泡状,部分细胞核固缩;SQ各组大鼠胰岛细胞受损程度较轻。结论 SQ能明显降低糖尿病大鼠的空腹血糖水平,提高血清C肽水平,保护胰岛细胞功能。

Abstract:Objective:To investigate the effects of SQ on fasting blood glucose(FBG),morpholog and function of islet cell in diabetic rats.Methods:SD diabetic models were established by streptozotocin(STZ) intraperitoneal injection.The experimental rats were divided into the following groups according to the means of intervention:model group,SQ low dose group,SQ middle dose group and SQ high dose group.The FBG and the weight were measured by every two weeks.At the end of 8 weeks of intervention,the blood was collected under an anaesthetic.Serum C-peptide levels were analyzed by ELISA.The pathology structures of pancreatic islets were observed by HE staining.Results:FBG in every group followed the order:model group(26.92±3.68) > SQ low dose group(23.87±2.86 > SQ high dose group(20.66±2.81) > SQ middle dose group(20.66±2.81) > control group(4.55±0.57).SQ middle and high dose groups had significant difference to model group(P<0.05).Serum C-peptide levels in every group followed the order:control group(2.27±0.47) > SQ middle dose group(1.76±0.91) > SQ high dose group(1.74±0.79) > SQ low dose group(1.11±0.36) > model group(0.70±0.62).SQ middle and high dose groups had significant difference compared to model group(P<0.05).The observation of histological examination revealed,in model group the cell amount of pancreatic islets decreased and coupled with volume increase,vacuolar cytoplast and disorder in rats.SQ groups’ pancreatic islet cells had mild damage in rats.Conclusion:SQ has effects on decreasing FBG of diabetic rats,increasing serum C-peptide levels,and protecting the islet cells.
