摘    要:采用杂交瘤技术将小鼠骨髓瘤SP_2/O细胞与用人甲胎蛋白抗原免疫的BALB/c小鼠脾细胞融合,经多次筛选,获得7株分泌抗人甲胎蛋白并对肝癌细胞有特异性亲和力的单克隆抗体杂交瘤细胞株,其培养液上清和腹水效价分别为10~(-2)和10~(-6~9-)。杂交瘤腹水经硫酸铵盐析,DEAE纤维素层析,或免疫吸附亲和层析法纯化,每ml腹水单克隆抗体的产量为1~2mg。用ELISA,免疫双扩散、聚丙烯酰胺凝胶电泳、间接免疫荧光法鉴定了单克隆抗体的特异性、亲和力、稳定性及IgG亚类。

Abstract:Seven mouse monoclonal antibodies to human alpha-fetoprotein (AFP) were produced by fusion of Sp2/O mouse myeloma cells with the spleen cells of BALB/c mouse immunized with AFP. The antibodies were isolated from ascitis fluid by ammonium sulfate precipitation and DEAE cellulose chromatography or immunoaf- finity chromatography. The purified monoclonal antibodies were shown to be present as one unique band by polyacryamid gel electrophoresis and to be bound specifically by SMMC-7721 hepatoma cell line. The results pave the way for subsequent studies of targeting cancer therapy.
