复合酶酶解法提取虎杖中白藜芦醇

摘    要:目的研究复合酶对虎杖中白藜芦醇的酶解法提取效果的影响。方法采用单因素实验及正交试验进行优选,考察酶解温度、酶解时间、复合酶用量等因素的影响,优化提取条件;采用HPLC法测定提解液中白藜芦醇含量,计算出提取率来比较提取效果。结果虎杖通过复合酶酶解后,能提高其白藜芦醇的提取率。结论较优工艺为复合酶用量5.0‰,40℃酶解36小时,该工艺对比直接提取法提取率提高近2倍,且重现性好,适宜工业化生产。

Abstract:Objective To study the effect of complex enzymes on extracting resveratrol from Polygonum cuspidatum. Methods The optimum extraction process was selected with single-factor experiment and orthogonal and the factors of enzymolysis temperature and time and complex enzymes dosage were monitored. Resveratrol extract was determined by HPLC, extraction yield was evaluated. Results The enzymolysis method can increase the extraction yield of resveratrol from Polygomun cuspidatum. Conclusion The optimum extraction process is as follows complex enzymes dosage 5.0‰, at 40℃ for 36 hours. The technology can improve the yield 2 times and it is suitable for industrial production.
