正交试验法优选小鼠四氯化碳矸损伤模型

摘    要:目的;采用正交试验法优选小鼠四氯化碳肝损伤最佳模型制作条件,方法:选择对小鼠血清转氨酶(ALT)有影响的四个因素(1)四氯化碳浓度;(2)肝毒剂中毒剂量;(3)中毒次数;(4)中毒途径,生个因素取3个水平,按L9(3^4)正交试验表安排是试验,以阳性药联苯双酯双小鼠四氯化碳肝损伤改善率为考察指标,评定四氯化碳中毒方案的优劣,结果:采用0.1%(v/v)的四氯化碳,按每10g小鼠0.2ml剂量腹腔一次注射的给药方案为佳,结论 以正交试验法优选出的小鼠四氯化碳肝损伤模型制作方案是可行的。

Abstract:Aim To explore the optimum method of CCl 4 injured liver model in mice by orthogonal test.Methods Four influential factors on transaminase in mice serum were chosen:(1)Concentration of CCl 4;(2)Dose of liver toxicosis;(3)Times of toxicosis;(4)Approaches of toxicosis,three levels of every factor.Arranged the tests based on L 9(3 4) orthogonal test table,improved ratio of Biphenyl dimethyl dicarboxylate (DDB) Tablet on CCl 4 injured liver in mice was regarded as detecting index,the optimum method of CCl 4 toxicosis was assessed.Results The optimum method was that:0.1%(v/v) CCl 4,0.2ml every 10g mice,ip,1time.Conclusion The injured liver model by CCl 4 selected by orthogonal test was feasible.
