9-顺-维甲酸对人肺鳞癌细胞分化和凋亡的影响

摘    要:目的研究9-顺-维甲酸(9-cis-RA)对人肺鳞癌L78细胞、A2细胞分化和凋亡的影响.方法以1和5μmol/L 9-cis-RA处理肿瘤细胞株,分别于0、12、24、36、48、72h检测对照组和用药组细胞数及细胞HE染色后的形态变化,应用流式细胞术分析细胞周期分布、Bcl-2表达和凋亡细胞数.结果9-cis-RA可抑制肺鳞癌细胞生长;用药后细胞分化征明显,可见凋亡细胞.流式细胞计数显示,用药组细胞G0/G1百分率明显升高,S期细胞百分率下降,细胞凋亡率增加,Bcl-2的表达下降.结论9-cis-RA能抑制人肺鳞癌L78细胞、A2细胞增殖,诱导细胞分化和凋亡.

Abstract:Objective:To investigate the effect of 9 cis RA on the differentiation and apoptosis of human lung squamous carcinoma cell lines(L78 and A2).Methods:Cell number and morphology were observed at 0,12,24,36,48,72h respectively after the treatment to the two study groups,the effect of 9 cis RA on the cell cycle and expression of Bcl 2 in L78 and A2 was also examined.Results:9 cis RA significantly decreased the number of L78 and A2 ( P<0.05 ,24h; P< 0.01,36h).Studies implicated G1 phage of the cell cycle was arrested,the percentage of G0/G1 phage increased( P< 0.01,22h),and the percentage of S phage cells decreased( P< 0.01,24h).Cell cycle arrest was associated with increased apoptosis and decreased expression of Bcl 2 protein.Conclusion:9 cis RA potently inhibited the growth of human squamous cancer cells in vitro,induced the differentiation and apoptosis of the cells.
