卵巢上皮性癌中Kras和P53的表达及临床意义

摘    要:目的:探讨Kras和P53蛋白在卵巢上皮性癌中的表达及与临床病理特征关系。方法:采用免疫组织化学SP法分别检测Kras、P53蛋白在非肿瘤性卵巢上皮组织10例及卵巢上皮性癌80例组织中的表达,并分析2种蛋白与临床病理特征的关系。结果:Kras蛋白在非肿瘤性卵巢组织中的阳性表达0例(0%),在上皮性癌中的阳性表达15例(18.75%)。P53蛋白定位于细胞核。P53蛋白在非肿瘤性卵巢组织阳性表达0例(0%),在上皮性癌组织中的阳性表达31例(38.75%)。(2)在卵巢上皮性癌中,Kras蛋白表达与卵巢上皮性癌组织分化程度、临床分期相关(P〈0.05),而与年龄、淋巴结转移无关;P53蛋白表达与组织分化程度、临床分期、淋巴结转移均相关,即P53蛋白在组织分化程度越低的上皮性卵巢癌中表达越高。结论:Kras和P53蛋白的表达与组织学分级相关,且两者高水平表达可能是卵巢癌预后不良的重要指标。

Abstract:Object ive: To investigate the expression and significance of Kras protein and P53 protein between normal ovarian tissue and epithelial ovarian carcinoma as well as their association with clinicopathologic features. Methods:90 cases were collected to study, including 80 cases with epithelial ovarian carcinoma and 10 cases of non-neoplastic ovarian tissue. Immunohistochemical methods were used to detect the expression of Kras and P53. Results:The positive expression rate of Kras between non-neoplastic ovarian tissue and epithelial ovarian carcinoma was 0.00% and 18.75%. The positive expression rate of P53 between non-neoplastic ovarian tissue and epithelial ovarian carcinoma was 0.00%and 38.75%.The expression of Kras in epithelial ovarian carcinoma was associated with the histologic grade amd the clinical stage, and not correlated with age and lymphatic metastasis. The expression of P53 in epithelial ovarian carcinoma was associated with the histologic grade, the clinical stage, and the lymphatic metastasis, and not correlated with age. Conclusions:The expression of Kras protein in low-grade epithelial ovarian carcinoma was higher than that in high-grade epithelial ovarian carcinoma and in the early stage, indicating that the role of Kras was important in the low-grade epithelial ovarian carcinoma. The expression of P53 protein in high-grade epithelial ovarian carcinoma was higher than that in low-grade epithelial ovarian carcinoma. The high expression of P53 protein usually was seen more in the late clinical stage,and associated with lymph node metastasis, which provided cogent evidence that the development of high-grade epithelial ovarian carcinoma was possibly related with P53 gene mutation.
